I have replaced with new parts
Igniters,wires, and PLUGS, breather hoses and rear connector for breather hoses. MAF sensor ,cam position sensor and all injectors. Found no intake leaks. Still only code is cylinder 5.

Vehicle: 2004 mercedes w211 e320 v6

One of the component that gets missed is the sparkplugs, if the sparkplugs is not secured at idle there is no misfire condition until it is on a cruise or highway condition. Check the sparkplugs swap it out with a different known cylinder,

The best way is to have the vehicle ignition spark tested, with a ignition analyzer. a lot of times the cause for this is the ECM. this is why a ignition analyzer is the best result. it can also determine if the cause is mechanical. MBZ there is no other way.
