I need help with a p0301 (cylinder 1 misfire)

I have a 2005 Pontiac Grand Am with the 3.4 v6. I have had a fluctuating, and difficult to diagnose misfire. I have concluded that it isn't the spark plug, wires, coil, ignition housing, air leak, or a blown head gasket. I tsted the injector for functioning by listening to it click with a screwdriver. I have also noticed that if i prime the car by holding the key on without starting it, several times, it can improve the misfire at idle and low speeds, up until about 50 mph. After a minute or two the misfire will go away for the day until the next cold start. Any help?

Swap out the ignition coil with a different coil and retest the system , if the misfire stays at the number 1 cylinder check the intake manifold gasket and see if there is any coolant seepage, or is there any coolant loss occurring the intake manifold are known to leak at both ends of the intake and create misfire condition
